چکیده

investigated the role of calpain in the endothelial dysfunction of Ang II. Objective: To demonstrate a mechanistic role for calpain in the endothelial dysfunction induced by Ang II/AT1R signaling. To establish endothelial-expressed calpains as an important target of AT1R signaling. Methods and Results: Subchronic administration of nonpressor doses of Ang II to rats and mice significantly increased vascular calpain activity via AT1R signaling. Intravital microscopy studies revealed that activation of vascular expressed calpains causes endothelial dysfunction with increased leukocyte–endothelium interactions

A novel role for calpain in the endothelial dysfunction induced by activation of angiotensin II type 1 receptor signaling.

RATIONALE The cytosolic protease calpain has been recently implicated in the vascular remodeling of angiotensin II (Ang II) type 1 receptor (AT(1)R) signaling. The role of Ang II/AT(1)R/calpain signaling on endothelial function, an important and early determinant of vascular pathology, remains though totally unknown. Accordingly, we investigated the role of calpain in the endothelial dysfunctio...

Combination Therapy with Losartan and α-Tocopherol in Acute Ureteral Obstruction-Induced Renal Excretory Dysfunction and Acidification Defect

Background: Previous study by the authors showed that a-tocopherol prevents oxidative stress but would not improve depressed excretory variables in post-obstructed kidney (POK) after release of 24-h unilateral ureteral obstruction (UUO). This study is a supplementary investigation on the effects of a-tocopherol combined with an antagonist of angiotensin-II type-1 (AT1) receptor on renal dysfunc...

Angiotensin II type 1 receptor blocker losartan attenuates locomotor, anxiety-like behavior and passive avoidance learning deficits in a sub-chronic stress model

Objective(s): Stress alters sensory and cognitive function in humans and animals. Angiotensin (AT) receptors have demonstrated well-established interactions in sets of physiological phenomena. AT1 receptors can play a part in stress-induced activation of hypothalamic-pituitary-adrenal (HPA) axis; besides angiotensinergic neurotransmission plays a pivotal role in stress-evoked physiological resp...

Endothelial Vasodilator Angiotensin Receptors are Changing in Mice with Ageing

Background: The vascular function of Angiotensin II-type-2 receptors in adults is controversial. We sought their location and function in mouse aortic rings at young and old mice. Materials and Methods: Male C57Bl mice (aged 4 and 14 months) were killed by CO2. The descending thoracic aorta was cleaned and dissected into rings. Aortic rings were mounted in Krebs’ solution at 37 °C an...

The roles of potassium channels in contractile response to urotensin-II in mercury chloride induced endothelial dysfunction in rat aorta

Urotensin-II (U-II), the most potent vasoconstrictor that has recently been recognized as a new candidate in cardiovascular dysfunction, might exert vasoconstriction through, at least partially, potassium channels that are predominant in both endothelial and vascular smooth muscle cells (VSMCs). The present study was designed to evaluate the roles of potassium channels in vascular responses to ...
